Big-spending Billericay will be hopeful of a large gate against the Shrimpers, who have tended to play a more youthful team in the competition.
Elsewhere, last year's finalists East Thurrock United visit Essex Senior League outfit Great Wakering Rovers, while Basildon United - quarter-finalists last term - will be hopeful of progression against league rivals Ilford.
Concord Rangers - three times winners between 2014 and 2016 - have been handed a home draw against Wivenhoe Town, and island rivals Canvey welcome either Saffron Walden Town or Waltham Forest.
Southend Manor have been handed a tricky-looking assignment at National League South powerhouses Braintree Town, and Bowers & Pitsea will travel to Heybridge Swifts - finalists two years ago and semi-finalists last time out.
The ties will all be played by November 11.
